Its expanded upon a little in the books what Greyscale's origins are. Or at least the myth. The Rhoyne that Tyrion mentioned is a GREAT river that cuts through western Essos and the people that lived on that river were the Rhoynar and they build a civilization that rivaled Valyria when the Dragons were still young. But Valyria hungered for more land, slaves, and riches as they grew and declaired several wars against the Rhoynar. In the last one they decimated the whole culture and the last of the Rhoynar fled but their last city's princes called upon the goddess of the river to strike down the Dragonlords. The banks of the river swelled and drowned many of the Valyrians but all those who survived and were touched by the water's mist were the first victims of Grayscale.
